Meredith Grey

Meredith narrates the show (with some exceptions), usually with voiceovers at the beginning and conclusion of the program.
Meredith is the daughter of Dr. Ellis Grey, who suffers from advanced Alzheimer's disease and was a close friend of Dr. Richard Webber, the chief of surgery at Seattle Grace Hospital.
She began a love affair with Dr. Derek Shepherd the night before she began her career as an intern, not knowing he was a surgeon at the hospital. She falls in love with him only to discover he is married when his wife Addison comes to join the staff at Seattle Grace.
She shares her house, which once belonged to her mother, with fellow intern Izzie. George used to live with her as well, but moved out following his disastrous one night stand with Meredith. A talented young doctor and a graduate of Dartmouth College, Meredith frequently battles self-doubt and mild depression over the respective situations with her mother and Derek.

Izzy
A statuesque blonde, Isobel "Izzie" Stevens put herself through medical school by modeling. Underwear. Wow.
When this fact is publicized by Dr. Alex Karev, who pinned pictures of her up in the locker room, Izzie flew off the handle. She later warmed up to Alex, however, and dated him briefly before catching him sleeping with a nurse and breaking it off. They remain friends, though.
At first, Izzie butted heads with Meredith over the latter's relationship with Dr. Shepherd. Izzie initially believed it was a plot for Dr. Grey to get ahead of her fellow interns, but the two have become good friends. They live in the same house, one that was also inhabited by George until the most recent episode.

Christina
She's abrasive, afraid of commitment, doesn't "do" emotion and is extremely competitive about getting the "cool" surgeries. She is of Korean descent and is a native of Beverly Hills, California. Cristina graduated from the Stanford University School of Medicine and maintains a love-hate relationship with her mother.
Despite being his polar opposite in many respects, she began dating Dr. Preston Burke during Season One. She became pregnant, only to have him break off their relationship for professional reasons.

George
When we first got to know Dr. O'Malley, he harbored a secret crush on Meredith, with whom he shares a house, from day one. George also has a host of other insecurities. His positive outlook on life, and the fact that he is more emotional than most men, often lead to conflict with other doctors.
George doesn't fit in with his own family, either. A Thanksgiving hunting trip resulted in a George unloading a lot of emotional baggage during a conversation with his blue-collar dad and brothers (one of whom later shot the dad, forcing George to operate).
A friend and confidant of Izzie, George won praise among the entire hospital staff for saving a man's life in an elevator during a power failure. With Dr. Burke talking him through it, George stopped the bleeding in a patient's heart single-handedly after Alex choked under pressure.
After great inner strife, he admitted his feelings for Meredith and slept with her in a moment of peak vulnerability for both. After some disastrous fallout, he moved out of her house and onto Dr. Burke's sofa. He remained there until Season Two came to a close - and he found a spot in bed next to his girlfriend, Callie Torres.

Alex
The arrogant, handsome intern who we first got to know as the man's man, both repulsive and irresistible to Izzie. The two had an on-again, off-again relationship that ended as Alex got caught sleeping with a nurse - in an attempt to regain confidence after "performance anxiety" problems with Izzie.
A former wrestler at the University of Iowa, he often tries to act more tough and aloof than he actually is. He and George O'Malley often clash due to their different personalities.
